Optimized Nuclei Preparation Workflow for Genomic Applications

The ability to accurately count the number of nuclei and intact cells is a crucial step in preparing for downstream applications. Researchers at Miltenyi Biotec performed nuclei counting on a flow cytometer as well as a DeNovix CellDrop Automated Cell Counter. Nuclei samples counted on the CellDrop (FL or FLi) were stained with Acridine Orange and Propidium Iodide (AO/PI) fluorescent dyes, allowing for clear differentiation between isolated nuclei, intact cells, and debris.

The nuclei count results were improved in accuracy by the CellDrop’s flexible protocol settings and robust analysis algorithm. The study concluded that the CellDrop’s count results were highly comparable to the Miltenyi Biotec MACSQuant™ Analyzer 10 Flow Cytometer, verifying the high accuracy of both methods.
